Thermo Scientific Pierce LC-SPDP is a long chain cross-linker that reacts with NHS esters and pyridyl disulfide groups The sexual group forms a cleavable (reducible) disulfide bond with the cysteine thiol group to realize the coupling of the amine and the thiol group.
Characteristics of LC-SPDP:
•
Reactive groups:
NHS Esterand
Pyridyl disulfide•
Can react with the following groups:Amino and thiol groups
•With free thiol groups Detectable by-products are released during the reaction; the reaction can be easily followed by measuring the release of 2-mercaptopyridine at 343 nm
•Disulfide bonds in the spacer arms can be treated with 10-50 mM DTT or TCEP (pH 8.5) Easy to cleave
•The spacer arm can also be easily cleaved using reducing SDS-PAGE loading buffer
•Cleatable cross-linker enables separation of cross-linked products
•Insoluble in water (first dissolve in DMF or DMSO)
•SPDP cross-linker is membrane permeable and cross-links within cells
•Comparable to many other SPDP-type reagents, including pegylated forms
Applications:• Reversible cross-linking of proteins
• Addition of sulfhydryl groups to proteins by attachment to lysine residues
• Formation of antibodies-enzymes through two-step reaction protein cross-linking and other bioconjugates
•Hapten attached to carrier protein for antibody production
•Preparation of immunotoxin conjugates
Specifications:We follow LC-SPDP is produced to higher specifications to produce highly specific bioconjugates, ensuring data integrity and providing a higher degree of consistency. Each batch of LC-SPDP is tested by quantitative NMR (the higher standard for cross-linker purity) to ensure purity >95%.
We offer three SPDP analogues: the standard form (SPDP), a longer spacer derivative (LC-SPDP) and a sulfonated water-soluble variant (
Sulfo-LC-SPDP). SPDP
( N-succinimide 3-(2-pyridyldithio)-propionate) and LC-SPDP (succinimide 6-(3-[2-pyridyl Dithio]-propionylamido)caproate) is a heterobifunctional, thiol-cleavable and membrane-permeable cross-linker. They each contain an amine-reactive
N-hydroxysuccinimide (NHS) ester that reacts with a lysine residue to form a stable amide bond. The other end of the spacer arm is terminated with a pyridine disulfide group, which reacts with a sulfhydryl group to form a reversible disulfide bond.
